Chymotrypsin for Autism Spectrum Disorders

This article will explore a new type of treatment for Autism Spectrum Disorders.

Autism is a difficult to treat condition characterized by repetitive behaviors, communicative problems, and inability to form appropriate social attachments. Autism is part of a spectrum of disorders which includes Asperger’s Disorder, and Pervasive Developmental Disorders. The incidence of these disorders may be as high as one in every 125 children. Up until several years ago there were zero drugs that were approved for the treatment of Autism. This is somewhat shocking when one considers that it is a fairly common condition. Recently 2 drugs were give the indication to treat Autism, Risperdal and Abilify. Both of these drugs are atypical antipsychotics which were originally designed to treat schizophrenia. They both carry the potential risks of Tardive Dyskinesia, EPS, Akathesia, sedation, Diabetes, and Hyperlipidemia / Metabolic Syndrome. Currently a new mechanism of treating Autism is being explored by Curemark Pharmaceutical in Rye, New York. They are looking at Chymotrypsin ( CM-AT ). Chymotrypsin is a digestive enzyme secreted by the pancreas. It is believed that children with Autism Spectrum Disorders produce low levels of this enzyme causing incomplete digestion of proteins. Chymotrypsin may help these children with Autism to digest proteins into essential amino acids necessary for the production of neurotransmitters and key brain signaling molecules. Some studies have shown that up to 70 % of Autism patients report GI digestive problems which may be due to low levels of chymotrypsin. Pancreatic enzymes have been prescribed to children for the treatment of cystic fibrosis, a genetic condition which causes pulmonary and digestive problems. Curemark is currently evaluating chymotrypsin at 12 trial sites in a phase 3 study of children age 3 to 8. So far no serious side effects have been reported which is very promising considering the potential risks associated with atypical antipsychotics for autism. Chymotrypsin also offers a unique mechanism of action which may help improve the core symptoms such as communicative problem, social deficits, and repetitive behaviors as well as the GI digestive problems. The product should be available in a sprinkle formulation that can be placed on food to help improve compliance. It is hoped that Chymotrypsin will offer new hope for these patients and their families. By Gregg L. Friedman MD, Hallandale Beach, Florida.
